Emirates will add a second Airbus A380 service between Dubai and Sydney on 2 June.
The airline currently operates the route with a Boeing 777-300ER, but it had already announced plans to upgrade the existing service to an A380.
Its two combined A380 flights will deliver an increase in capacity of 1,890 seats per week.
Emirates and Qantas will launch a strategic partnership on 31 March, replacing the Australian carrier's earlier arrangement with British Airways.
The two airlines will jointly offer 98 weekly services between Australia and Dubai, including five daily A380 flights from 2 June.
