Emirates will start daily Airbus A380 services between Dubai and Barcelona on 1 February.
The airline currently runs a Boeing 777-300ER operation linking its base with the Catalan city.
Configured with 517 seats - including 14 in first class and 76 in business - the A380 will boost capacity on the route by 2,198 seats per week, says Emirates.
The carrier flies A380s to 21 destinations and will add four more over the next six months: Brisbane on 1 October, Los Angeles on 2 December, Mauritius on 16 December and Zurich on 1 January.
Emirates' fleet Includes 35 A380s, and it has a further 55 on order.
