Qantas will launch a direct Sydney-Gladstone service on 4 March 2013.
The carrier will operate eight times weekly on the route, using QantasLink's Bombardier Dash 8 Q400 aircraft.
The new route is intended to satisfy growing corporate demand for direct services between the two cities, Qantas's domestic chief executive Lyell Strambi said in a statement.
In addition, Qantas will increase seat capacity on four of its domestic routes.
On the Dubbo-Sydney route, the airline will replace the Q300 aircraft operating on the route with a Q400. This will add 384 seats per week to the eight times weekly route.
It will also replace the Q300 used on the thrice-weekly service on the Port Macquarie-Sydney route with a Q400.
